BASIS OF KINETIC ANALYSIS OF THE MULTI-SITE ENZYME SYSTEMS
Synopsis
At the beginning of the last century, the kinetic theory of single-site enzyme systems was developed and the methodology of their complete kinetic analysis was refined. Its essence consisted in the transformation of the hyperbolic function into a linear dependence, which is characteristic only for single-site systems. But, later it was found that many enzymes, due to multisite, are characterized by curvilinear kinetic dependence. It became necessary to develop principles of kinetic analysis of such curves and new kinetic parameters. Despite many attempts, this goal could not be achieved. Z. Kometian's monograph presents for the first time the theoretical foundations of the complete deciphering of the molecular mechanism of the multisite enzyme system and the statistically reliable methods of determining the main kinetic parameters based on experimental data.
